Abstract

Background: Cyclooxygenase-2 (COX-2) is an enzyme responsible for the conversion of arachidonic acid into prostaglandins. Despite previous studies, there is controversy considering the exact role of COX-2 in the pathogenesis of type 2 diabetes mellitus (T2DM) and its related adverse cardiovascular events. We aimed to examine the association between COX-2 rs20417 genetic polymorphism with increasing susceptibility to T2DM and whether it enhances the development of cardiovascular complications in T2DM. Methods: A total of 244 individuals were recruited, including 164 T2DM patients (further subdivided into uncomplicated and cardio-complicated cases) and 80 healthy control subjects. The different genotypes for the COX-2 rs20417 polymorphism were assayed through polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis. Lipid profile and oxidative stress markers (oxidized LDL (OX-LDL) and malonaldehyde (MDA)) were assayed. Results: The CC, GC, and GC+CC variants of the COX-2 rs20417 polymorphism were significantly more prevalent in T2DM group compared with the healthy control group (P
